Entrectinib, a potent, CNS-active, TRK and ROS1 inhibitor is associated with occurrence of bone fractures, particularly in pediatric patients. We embarked on computer-guided and experimental investigations to identify the underlying mechanism that should help safe dosing of children receiving this drug. An artificial intelligence (AI)-based approach was used to allow for hypothesis generation regarding drug effects in a clinic...

Beech trees form major parts of lowland temperate forests in the Western Palearctic. To protect biodiversity, many beech forests have been designated as World Heritage Sites or established as networks of beech forest reserves. However, the insect communities within these protected forests have not been well explored. In this study, elaterids (Elateridae, Coleoptera) in 26 beech forests, from France in the west to Iran in the e...

Exposure to multiple substances is a challenge for risk evaluation. Currently, there is an ongoing debate if generic “mixture assessment/allocation factors” (MAF) should be introduced to increase public health protection. Here, we explore concepts of mixture toxicity and the potential influence of mixture regulation concepts for human health protection. Based on this analysis, we provide recommendations for research and risk a...

Key message Genotype-by-environment interactions of secondary traits based on high-throughput field phenotyping are less complex than those of target traits, allowing for a phenomic selection in unreplicated early generation trials. Abstract Traditionally, breeders’ selection decisions in early generations are largely based on visual observations in the field. With the advent of affordable genome sequencing and high-throughput...

A variety of human activities have been identified as driving factors for the release and spread of invasive earthworm species in North America. Population genetic markers can help to identify locally relevant anthropogenic vectors and provide insights into the processes of population dispersal and establishment. We sampled the invasive European earthworm species Lumbricus terrestris at nine sites and several bait shops within...

Invasive infections caused by carbapenemase-producing bacteria are associated with excess mortality. We applied a rapid diagnostic test (RDT) on clinical samples with an elevated likelihood of carbapenemase-producing bacteria and documented its impact on antibiotic treatment decisions. Among 38 patients, twelve tested positive for infections caused by carbapenemase-producing bacteria (31.6%), mainly in blood cultures. KPC ( n...

Shifts from coral to algal dominance are expected to increase in tropical coral reefs as a result of anthropogenic disturbances. The consequences for key ecosystem functions such as primary productivity, calcification, and nutrient recycling are poorly understood, particularly under changing environmental conditions. We used a novel in situ incubation approach to compare functions of coral‐ and algae‐dominated communities in t...
